研究概要

有用な生物活性を示す低分子化合物のはたらきを分子レベルで理解するためには、まず生体内あるいは細胞内における標的分子を知ることが鍵となります。生物活性分子の代表的な標的分子はタンパク質です。生物活性分子は特定のタンパク質に対して結合相互作用を起こし、タンパク質の本来の機能に大きな変化をもたらすことでその生物活性を発現すると考えられています。
 私たちの研究グループでは、作用機構の解明に向けて、有機合成法を用いて生物活性分子の誘導化を行い、結合タンパク質を効率的に探索するための機能を有したプローブを開発しています。生体内や細胞内の多数のタンパク質群の中から特異的な結合タンパク質を発見するためには、探索の網羅性が高く、生理条件下で高感度かつ高選択的に結合タンパク質を検出し分離する技術が必要となります。そこでフォトアフィニティーラベリング法とよばれる低分子―タンパク質結合複合体の架橋法に着目し、望みの結合タンパク質の選択的な捕捉と同定を可能とする一般的な方法論の実現に向けて、以下の研究プロジェクトを推進しています。

1. 活性型/不活性型2種プローブフォトアフィニティーラベリング法の開発
2. 抗癌活性化合物OSW-1を基盤としたフォトアフィニティープローブの開発と結合タンパク質の探索

Summary

Identification of binding proteins is critical for understanding the mechanism of action for bioactive small-molecules. Certain small-molecules are known to display their biological activity by first binding to specific proteins, which results in altering their natural functions.
     Our research group is interested in developing new chemical probes based on bioactive small-molecules to facilitate efficient exploration of their specific binding proteins. In order to rapidly discover binding proteins out of complex mixtures of cellular proteins, chemical probes need to be capable of selective and sensitive detection of the desired proteins under the physiological condition. To address these issues, we employ photoaffinity labeling, which allows covalent crosslinking of small molecule-protein bound complexes. Some of the currently ongoing research projects are listed as below.

1. Development of active/inactive dual-probe photoaffinity labeling methods
2. Development of photoaffinity probes based on an antitumor agent OSW-1 and their application to the target identification studies
